چکیده

The use of mutagens in plant breeding is used to create new germplasm, increase agricultural yield, quality, and resistance diseases pests. Mutagens are physical or chemical factors that can alter the DNA RNA structure an organism, causing mutations above expected level. One most common potent EMS (ethyl-methane sulfonate), which produces point plants, but a lesser degree also cause loss deletion chromosomal region. This study inter-primer binding site (iPBS) coupled restriction enzyme digestion (CRED-iPBS) technique analysis determine effect on methylation rates wheat genotypes at seedling growth stage. Treatments with five different concentrations (0%; control, 0.1%, 0.2%, 0.3%, 0.4%) four times (0; 3, 6, 9 h) were used. Inter-primer markers employed assess genomic instability cytosine treated wheat. In seeds times, disappearance regular bands formation due effects mutagen revealed genetic diversity exists. CRED-iPBS 3 h + 0.1% treatment produced highest MspI polymorphism value (19.60%), while lowest (10.90%). mutagenic treatments had considerable variety impacts According current research, mutagenesis may be practical method for accelerating programs produce enough populations. Mutation-assisted subsequent selection desirable mutants using carried out utilizing integrated strategy.

Salt-Induced Tissue-Specific Cytosine Methylation Downregulates Expression of HKT Genes in Contrasting Wheat (Triticum aestivum L.) Genotypes

Plants have evolved several strategies, including regulation of genes through epigenetic modifications, to cope with environmental stresses. DNA methylation is dynamically regulated through the methylation and demethylation of cytosine in response to environmental perturbations. High-affinity potassium transporters (HKTs) have accounted for the homeostasis of sodium and potassium ions in plants...

Potassium induced salinity tolerance in wheat (Triticum aestivum L.)

Water culture experiments were conducted to study the response of ten wheat genotypes to external K application (10 mmol KCI dm3) at seedling stage under saline condition (0 and 100 mmol NaCl dm3). The data showed that there was an increase in the shoot and root length with the application of external K. The increase was more pronounced under control than under saline conditions. The better per...

The wheat (Triticum aestivum L.) leaf proteome.

The wheat leaf proteome was mapped and partially characterized to function as a comparative template for future wheat research. In total, 404 proteins were visualized, and 277 of these were selected for analysis based on reproducibility and relative quantity. Using a combination of protein and expressed sequence tag database searching, 142 proteins were putatively identified with an identificat...

Ethyl Methanesulfonate (EMS)-Mediated Mutagenesis of Cucumber (Cucumis sativus L.)

Ethyl methanesulfonate (EMS) is a stable and effective chemical mutagen. In this study, cucumber (Cucumis sativus L. cv. “Shannong No. 5”) seeds were treated by 1% EMS for 12 h, 24 h and 48 h to optimize EMS mutagenesis and determined median lethal dose of EMS (1% EMS and 24 h) for “Shannong No. 5”. After treated by 1% EMS for 24 h, 541 M1 plants were grown in greenhouse for phenotype investiga...

Simulation of Maintenance Respiration in Wheat (Triticum aestivum L.)

Respiration has a profound influence on crop yield because dry matter accumulation by a crop is closely related to assimilation of CO and respiratory activity. This paper describes the development of a 2 simulation model for respiration in wheat. While the growth respiration can be assumed to be proportional to the assimilation of CO , the model concentrated on the effects of biotic and environ...
